The basic steps in obtaining a gene from any organism, making recombinant DNA and then cloning this gene in a bacteria are described in this practical class. IC-PBL 수업 문제 If a particular gene is big in size and it is not available commercially, How do you clone a gene and use it in your research - Not all genes and its expression plasmids are available commercially. If we need to analyze the functions of a novel gene we have to clone it from the cDNA and subclone it into a expression vector. In this class student can learn how to clone a gene from cDNA. Additionally, if the gene is very big in size, it is difficult to clone. So students will learn how to solve the problem cloning a big size genes into expression vector and express it in a mammalian cell lines. OR Not all genes and its expression plasmids are available commercially. Additionally, if the gene is very big in size, it is difficult to clone. If the gene is having highly GC content, students learnt how to solve cloning by adding GC melt, DMSO while performing PCR. Additionally, students learnt how to solve the problem cloning a big size genes by improving ligation efficiency into expression vector and transformation efficiency in bacterial system.
